Screening for Genes Associated with Motility in Malignant Glioma Cell Lines

Abstract
Objective: The most characteristic feature of malignant glioma is an early and extensive infiltration into the adjacent parenchymal structures. The failure of surgical treatment in glioma is mainly due to invasion of the brain by tumor cells beyond resection areas. We focused on detecting the possible expression changes as the determining factors for malignant glioma¡¯s motile ability.

Materials and Methods: We compared the motility of three malignant cell lines(U87MG, U343MG-A, U87MG-10¡¯), using simple scratch test. Based on the results of GenefishingTM Differentially Expressed Gene(DEG) technique, we formed 39 motility-associated genes that differentially displayed between motile and non-motile cell lines.

Results: We confirmed 5 of 39 genes that showed different expressions on differential display polymerase chain reaction( DD-PCR) by reverse transcription polymerase chain reaction(RT-PCR) and Northern blot.

Conclusion: Our study shows that these genes may act as possible determining factors of high motility in the malignant glioma cell lines. These findings can help us understand the molecular invasion mechanism in gliomas.
